The hierarchical structure of chromatin involves ordered assembly of nucleosomes and folding of the nucleosomes into densely packed states. These processes require the participation of histone chaperones, nucleosome remodeling complexes, and additional structural proteins. We have used a combination of biochemical, structural and biophysical approaches to investigate the molecular mechanisms of chaperone-histone and protein-nucleosome interactions, and have determined the architecture of the 30nm chromatin fiber. Our thematic investigations should reveal underlying principles of chromatin folding and dynamics, as well as providing mechanistic insights into epigenetic regulation of gene expression.
