Mitochondria are iconic structures in biology. In addition to being the prime location for ATP production, as indicated by their well-known moniker “powerhouses of the cell”, mitochondria are morphologically dynamic organelles that possess highly varied proteome landscapes and home to a vast array of metabolic and signaling processes. Mitochondrial dysfunction has been profoundly implicated in major human diseases, including cancer, diabetes, and neurodegenerative diseases, and in the normal aging process. How mitochondrial dysfunction arises and contributes to often tissue- and cell type- specific disease pathology is poorly understood. Autophagy, or self-eating, is an evolutionarily conserved biological process that helps cells survive harsh, nutrient-limiting conditions. Defects in this process have also been linked to major human diseases. Many important questions regarding the role of autophagy in human diseases remain to be answered. Recent studies by our lab and others have revealed that certain neurons rely heavily on the autophagy pathway to remove damaged mitochondria, and that defect in this clearance process called mitophagy contributes to Parkinson’s disease. In my talk, I will discuss our most recent findings on the regulation and function of mitophagy in the contexts of neuromuscular degeneration and cancer biology, and our new understanding of mitophagy as an integral part of a mitochondrial quality control continuum.
